Will the item gain more or less if you do all 100 levels in one trip? or is it better to stop after each item general/king you kill?

It's best to stop if you plan to max the weapon out. And it may also be a good idea to stop and save every once in a while anyway.

If you're trying to max out the weapon, you're going to want to stop and save very often until you get the bonus 100 levels. Additionally, you're going to want to stop every time you kill a king (Using a Gency after the first kill, then exiting the stage after the second) or god (Only on the first Gency kill) to put in a specialist for the stat bonus.

But if you're not planning on capping the weapon at level 200 with max specialists, then it's really a matter of what you feel makes it easier to get through the item.

whats this about puttin a specialist in for a stat bonus?

and really what i wuz askin wuz.. does killing an item general/king/god bring the stats up faster? like if i kill a general, does the item get a bonus per level or just an added bonus at the end of all the levels?

Killing Generals/Kings/Gods gives you an additional stat bonus for each stat specialist you have on an item. So if you wanted to maximize a weapon's attack growth, you'd put as many Gladiators as you could on the weapon. And the General/King/God bonus isn't cumulative. It gives just as much for one floor clear as it does for all 10.
